Dynamic Adoption of Anergy by Antigen-Exhausted CD4+ T Cells

Abstract Exhausted immune responses to chronic diseases represent a major challenge global health. To explore their cell fate and plasticity in exhaustion, we analyzed CD4+T cells mouse model with regulatable antigen presentation. When the are driven through effector phase, then exposed different levels of persistent antigen, they lose Th1 functions, upregulate exhaustion markers, resemble naturally anergic become unable help B and, at highest dose, undergo apoptosis. Mechanistically, TCR signaling pathways modulated increasing dose time. Ca2+ fluxes were found be more robust presentation while MAPK Akt easily desensitized. Transcriptomic analyses show dynamic adjustment gene expression accumulation signals over period weeks. Upon removal, recover functionality losing anergy markers. Preliminary data indicate that antigen-exhausted retain for least 30 days able adjust antigen. Our suggest response CD4+ T persisting contribute better understanding disease.
